Kinobeads™ Screening
Using Kinobeads™ we can screen libraries of up to 100,000 compounds against native kinases directly in lysates of cells.
In lead optimization, we use Kinobeads™ as a primary assay system to drive optimization of the binding potency of candidate molecules towards the target, and selectivity over other, related kinases.
The assay principle is the same as for » profiling experiments where the Kinobeads™ compete with the test compound for binding to the target. The kinases captured on the beads are then quantified by spotting on a reversed phase microarray and using immunodetection. By spotting several microarrays from a single sample, multiple targets can be detected at the same time.
This provides a 'molecular high content' screening approach, where, for example, all kinases of a particular pathway can be screened in parallel.
