Knowing all the components of
a disease pathway allows us to select the best target candidates - by the extent of chemical tractability and position in the network - for progression as drug discovery opportunities.

Case study
We have mapped the Tumor Necrosis Factor α (TNFα) signalling pathway. TNFα is a cytokine that triggers a key pro-inflammatory signalling pathway. We mapped the protein interaction network underlying this disease pathway, which resulted in a landmark publica- tion as the first physical and functional map of a human disease pathway (» Bouwmeester et al. Nature Cell Biology 6, 97-105 2004). Several interactors were found to modulate the pro-inflammatory response and therefore represent novel opportunities for therapeutic intervention in diseases, like rheumatoid arthritis and psoriasis, where inflammatory processes play a major role.
